OGMP- DNA Sequencing strategy
DNA sequencing is performed by the dideoxy chain termination method
(Sanger et al, 1977). Cycle-sequencing reactions with plasmid DNA are
performed for Li-Cor sequencing, while standard reactions with T7 DNApol,
35S-dATP and single-stranded DNA templates are performed for clone "fingerprinting", which consists of only an A-reaction, instead of
GATC-reactions.
