YBDG Medium for Amoebidium (biotin added)


For one liter medium:
                                 
     brain heart infusion (Difco)          3,7 g
     tryptone                              5   g
     yeast extract                         5   g
     K2HPO4                                0.3 g
     KH2PO4                                0.3 g
     glycerol                              1 %

     (pH comes out to about 6.8)

     For solid media add 1,7 % Agar

     Autoclave 

Add 2% Glucose, 2 mg thiamine and 0.05 mg biotin from 
sterile 50% stock, before use.  


Amoebidium is a slow grower, and it is important to start well-grown starter cultures before going to larger volumes. For liquid medium use about 200 mL in a 2L Erlenmeyer flasks, inoculate with cells from petri dish, and incubate at room temperature (slow shaking), for several weeks.
